The first total synthesis of one of the spicamycin congeners, SPM VIII (3), is described. A preliminary model study for construction of the characteristic N-glycoside linkage in spicamycin using tetra-O-benzyl-beta-D-mannopyranosylamine (13) and halopurines 5 revealed that Pd-catalyzed conditions successfully provided the coupling products 14 and 15 in good yields. It was also shown that thermal anomerization of the N-glycosides easily occurred, which resulted in the predominant formation of the beta-anomer as the thermodynamically favored compound, and the activation energy of anomerization of 15 was estimated to be ca. 30 kcal/mol. The novel aminoheptose unit of spicamycin 6 was prepared stereoselectively by carbon elongation of an acyclic aldehyde, prepared by ring cleavage reaction of a highly functionalized cyclohexane derived from naturally abundant myo-inositol. The Pd-catalyzed coupling reaction of the beta-heptopyranosylamine 6 with protected 6-chloropurine 5d, followed by deprotection, provided spicamycin amino nucleoside 2, whose condensation with dodecanoylglycine completed the total synthesis of 3. This study confirmed the proposed unique structure of a novel nucleoside antibiotic.  相似文献

Five new ent-kaurane-type diterpenoids and a new gymnomitrane (=barbatane)-type sesquiterpenoid have been isolated from the Japanese liverwort Jungermannia truncata NEES, together with twelve previously known ent-kaurane-type diterpenoids. The structures of the new compounds were elucidated by two-dimensional (2D) NMR experiments and chemical reaction. Some of the isolated compounds showed cytotoxicity against human leukemia cell lines and induced apoptosis.  相似文献

Systematic synthesis routes have been developed for the linear-shaped rhenium(I) oligomers and polymers bridged with bidentate phosphorus ligands, [Re(N--N)(CO)3-PP-{Re(N--N)(CO)2-PP-}(n)Re(N--N)(CO)3](PF6)(n+2) (N--N = diimine, PP = bidentate phosphine, n = 0-18). These were isolated by size exclusion chromatography (SEC) and identified by (1)H NMR, IR, electrospray ionization Fourier transform mass spectrometry, analytical SEC, and elemental analysis. Crystal structures of [Re(bpy)(CO)3-Ph2PC[triple bond]CPPh2-Re(bpy)(CO)3](PF6)2, [Re(bpy)(CO)3-Ph2PC[triple bond]CPPh2-Re(bpy)(CO)2-Ph2PC[triple bond]CPPh2-Re(bpy)(CO)3](PF6)3 and [Re(bpy)(CO)3-Ph2PC2H4PPh2-{Re(bpy)(CO)2Ph2PC2H4PPh2-}(n)Re(bpy)(CO)3](PF6)(n+2) (bpy = 2,2'-bipyridine, n = 1, 2) were obtained, showing that they have interligand pi-pi interaction between the bpy ligand and the phenyl groups on the phosphorus ligand. All of the oligomers and polymers synthesized were emissive at room temperature in solution. For the dimers, broad emission was observed with a maximum at 523-545 nm, from the (3)MLCT excited-state of the tricarbonyl complex unit, [Re(N--N)(CO)3-PP-]. Emission from the longer oligomers and polymers with > or = 3 Re(I) units was observed at wavelengths 50-60 nm longer than those of the corresponding dimers. This fact and the emission decay results clearly show that energy transfer from the edge unit to the interior unit occurs with a rate constant of (0.9 x 10(8))-(2.5 x 10(8)) s(-1). The efficient energy transfer and the smaller exclusive volume of the longer Re(I) polymers indicated intermolecular aggregation for these polymers in an MeCN solution.  相似文献

A system to perform post-column derivatization capillary electrochromatography (CEC) was developed for the first time. The system mainly included a 4-microm (O.D.) silica packed column (200 mm effective length x 0.1 mm inner diameter I.D.) with micro-magnetic particles (MMPs) frits, a T-junction connector, an in-line fluorescence detector and a high-voltage power supply. The system was evaluated by using histamine (HA) as a standard biogenic amine for this study. A 5 microM HA solution was loaded at the anodic site of the capillary column by applying 3 kV for 5s. Then, HA was electrophoretically eluted with a 20mM phosphate buffer (pH 7) by applying 3 kV, and was derivatized with 3mM o-phthalaldehyde (OPA)/N-acetylcysteine (NAC) in 100 mM borate (pH 10), which was continuously delivered through the reagent-loading capillary tube by gravity into the T-junction connector. HA derivative was finally detected with the in-line fluorescence detector (lambda(Ex)=340 nm, lambda(Em)=450 nm) at 9.7 min after sample loading. To test the utility of this system, it was next employed for its ability to detect the presence of HA and other kinds of biogenic amines, including cadaverine (Cad), spermidine (Spm) and tyramine (Tyr) in tuna-meat, once the validity of the method had been confirmed.  相似文献

Three new lanostane‐type triterpenoids, inonotsutriols A ( 1 ), B ( 2 ), and C ( 3 ) were isolated from the sclerotia of Inonotus obliquus (Pers .: Fr.) (Japanese name: kabanoanatake; Russian name: chaga). Their structures were determined to be (3β,21R,24S)‐21,24‐cyclolanost‐8‐ene‐3,21,25‐triol ( 1 ), (3β,21R,24R)‐21,24‐cyclolanost‐8‐ene‐3,21,25‐triol ( 2 ), and (3β,21R,24S)‐21,24‐cyclolanosta‐7,9(11)‐diene‐3,21,25‐triol ( 3 ) on the basis of NMR spectroscopy including 1D and 2D experiments (1H,1H‐COSY, NOESY, HMQC, and HMBC) and EI‐MS.  相似文献

The complexation of Keggin-type polyoxometalate [alpha-SiW 12O 40] (4-), macrocation [Cr 3O(OOCC 2H 5) 6(H 2O) 3] (+), and monovalent cation A (+) forms ionic crystals of A 2[Cr 3O(OOCC 2H 5) 6(H 2O) 3] 2[alpha-SiW 12O 40]. nH 2O [A = Na ( 1a), K ( 2a), Rb ( 3a), NH 4 ( 4a), Cs ( 5a), and tetramethylammonium (TMA) ( 6a)]. Single crystal (1a- 4a and 6a) and powder (5a) X-ray analyses have shown that the ionic crystals possess 2D layers of polyoxometalates and macrocations. Compounds 2a- 5a had almost the same structure, while the layers in 1a and 6a stack in different ways. The structures and sorption properties of 2b- 5b are investigated in more detail. The interlayer distances of guest free phases 2b- 5b increase with the increase in the ionic radii of the monovalent cations, which reside between the layers. Compounds 2b- 5b possess hydrophobic and hydrophilic channels, which exist between the layers and through the layers, respectively. The volumes of the hydrophobic channels increase in the order of 2b < 3b approximately 4b < 5b, and those of the hydrophilic channels increase in the order of 2b < or = 3b < or = 4b < 5b. Single-crystal X-ray structure analyses of 2a- 4a have shown that the water of crystallization resides in the hydrophilic channel. It is probable that the water of crystallization in 5a resides in the hydrophilic channel in the same manner as those in 2a- 4a since 2a- 5a have almost the same structure. The water vapor sorption profiles of 2b- 5b are approximately reproduced by a linear driving force model. Therefore, water molecules sorbed in 2b- 5b probably reside in the hydrophilic channel. The n-propanol sorption profiles are reproduced by the summation of the linear driving force model, showing that two independent barriers exist in the n-propanol sorption. The in situ IR spectra of n-propanol sorbed showed the presence of two n-propanol species. These data show that n-propanol is sorbed into both hydrophilic and hydrophobic channels. Compound 5b sorbs halocarbons in the hydrophobic channel, while 2b- 4b exclude them.  相似文献

Humic substances that preferentially adsorb at the air/water interfaces of water or aerosols consist of both fulvic and humic acid. To investigate the chemical reactivity for the heterogeneous reaction of gaseous ozone, O(3)(g), with aqueous iodide, I(-)(aq), in the presence of standard fulvic acid, humic acid, or alcohol, cavity ring-down spectroscopy was used to detect gaseous products, iodine, I(2)(g) and an iodine monoxide radical, IO(g). Fulvic acid enhanced the I(2)(g) production yield, but not the IO(g) yield. Humic acid, n-hexanol, n-heptanol, and n-octanol did not affect the yields of I(2)(g) or IO(g). We can infer that the carboxylic group contained in fulvic acid promotes the I(2)(g) emission by supplying the requisite interfacial protons more efficiently than water on its surface.  相似文献

The oxygen partial pressure (P(O2)) dependence of in situ X-ray absorption spectroscopy (XAS) at the Co and Fe K edges was measured simultaneously and continuously at 900 and 1000 K. These experiments, which were performed during reduction, changing P(O2) from 1 to 10?4 atm, were used to investigate each valence related to Co and Fe in (La0.6Sr0.4)(Co0.2Fe0.8)O3?δ (LSCF). The absorption edge shift of the Co K edge was more than twice that of the Fe K edge at 1000 K during reduction. For quantitative analysis, X-ray absorption near-edge structure spectroscopy was carried out at the Co and Fe K edges; the results indicated that the Co valence decreased more easily than the Fe valence; that is, the oxygen preferentially left from the oxygen sites around Co.  相似文献
